Description: IKKbeta (IKKβ) (also known as IKK2) is a Ser/Thr kinase which phosphorylates IkappaB protein substrates. In unstimulated cells, IKK2/β remains as an inactive kinase as a complex with IKK1/α and IKKg (also known as NEMO). In response to a large number of stimiuli including cytokines, IKK2/b is activated which in turn phosphorylates IkB protein substrates (IkBα, IkBβ, IkBe and p105). Phospho-IkB proteins are rapidly ubiquitinated and degraded by the proteasome liberating NF-kB transcription factors to bind their target DNA sequences. The commonly used substrate for in vitro phosphorylation is GST-IkBa 1-54 where Ser32 and Ser36 of IkBa undergo phosphorylation. A large number of other proteins including p53 also undergo phosphorylation by IKK2/β. IKK2/β is active even when IKK1/2 is absent in cells. Recombinant IKK2/β can phosphorylate its target substrates in the absence of both IKK1/a and NEMO. The truncated IKKβ (His-IKKb 1-700) can interact with IKKα but not NEMO. IKK2/β is hyperactive in many inflammatory diseases and cancer.
Composition: 0.5 mg/ml in 25 mM pH 8.0 Tris-HCl, 150 mM NaCl, 1 mM EDTA, 1 mM DTT and 50% Glycerol.
