Thermolabile UDG (UNG) Uracil-DNA- Glycosylase
Uracil-DNA glycosylase (UDG or UNG) hydrolyses single or double-stranded DNA containing dUTPs. Consequently, if PCRs are performed in the presence of dUTPand dNTPs mixtures, a UNG step before each run will eliminate undesired PCR products from the previous runs.
Source: Thermolabile UDG expressed in and purified from E.coli
Storage conditions: For long term storage the UDG should be stored at -20°C in a constant temperature freezer. When stored under these conditions the reagents are stable for at least two months. Avoid repetitive freeze-thaw cycles.
Protocol: If UDG is required, please add an initial step before the usual cycling protocol: 5 min at 25°C.
Storage Buffer: 20 mM Tris pH 7.5, 200 mM NaCl, 0.1 mM EDTA, 10 mM DTT, 50% Glycerol.
Procedure: Add UDG to a final concentration of 0.05 U/uL (1μl of 1 u/μL per 20 μL) reactionwhen using any PCR or qPCR kits containing dUTP.
Unit Definition: One unit is defined as the amount of enzyme that catalyses complete degradation of 100 ng of uracil containing double stranded DNA in 5 min at 25°C.
